Overview

MBD2 shRNA is a short hairpin RNA designed to silence the expression of Methyl-CpG-binding domain protein 2 (MBD2). MBD2 is a key component of the Nucleosome Remodeling and Deacetylase (NuRD) co-repressor complex, which plays a critical role in epigenetic regulation and gene silencing. In the context of Acute Myeloid Leukemia (AML), research has explored the knockdown of MBD2 to understand its role in tumor initiation and sensitivity to chemotherapy. Studies indicate that MBD2 and MBD3 may have redundant functions within the NuRD complex in AML cells, as significant phenotypic changes—such as reduced colony formation or increased sensitivity to cytarabine (AraC)—often require the simultaneous disruption of both MBD2 and MBD3 pathways. This research has been primarily conducted in academic settings, such as Virginia Commonwealth University, to identify potential new therapeutic targets for AML.
