Obsidian Therapeutics is developing tumor-infiltrating lymphocytes (TILs) engineered to express a membrane-bound form of interleukin-12 (mbIL12) using its proprietary cytoDRiVE platform. This technology enables the pharmacological regulation of mbIL12 expression, allowing for precise spatiotemporal control over cytokine activity within the tumor microenvironment. By anchoring IL-12 to the cell membrane and making its expression regulatable by a small molecule, the therapy aims to enhance the anti-tumor potency of TILs and overcome immunosuppressive signals in solid tumors while avoiding the severe systemic toxicities typically associated with recombinant IL-12 administration. Preclinical studies in xenograft and syngeneic solid tumor models have demonstrated that these engineered cells can drive the regulation of pharmacodynamic markers and improve tumor control.
