Drug intelligence / Profile preview
mbIL15-BCMA CAR-T is an investigational, autologous chimeric antigen receptor T-cell (CAR-T) therapy engineered to target B-cell maturation antigen (BCMA) and co-express a pharmacologically regulated membrane-bound interleukin-15 (mbIL15). Developed by Obsidian Therapeutics using its proprietary cytoDRiVE™ platform, the therapy features mbIL15 fused to a carbonic anhydrase 2 (CA2) drug responsive domain (DRD). This design allows for the precise, titratable control of mbIL15 expression on the cell surface via the administration of the FDA-approved small molecule ligand acetazolamide (ACZ). The regulated expression of mbIL15 is intended to promote antigen-independent CAR-T cell survival, expansion, and persistence, as well as the development of a memory stem cell phenotype, thereby enhancing anti-tumor efficacy and safety in patients with multiple myeloma.
