Overview

mbIL21-expanded natural killer cells is an investigational adoptive cell therapy consisting of haploidentical natural killer (NK) cells expanded ex vivo using the FC21 platform. This platform utilizes K562 feeder cells genetically modified to express membrane-bound interleukin-21 (mbIL-21) and 4-1BB ligand (CD137L), which promotes significant expansion and enhances the anti-tumor cytotoxicity of the NK cells. Developed by the Hospital de Clínicas de Porto Alegre, this therapy is being evaluated for the treatment of relapsed or refractory acute myeloid leukemia (AML). The treatment protocol typically involves lymphodepleting chemotherapy with fludarabine and cytarabine (FLAG regimen) followed by multiple infusions of the expanded NK cells. This technology was later licensed to Kiadis Pharma (now part of Sanofi) for further development as K-NK002.
