MBL949 is a half-life extended recombinant human Growth Differentiation Factor 15 (GDF15) dimer developed as a long-acting GDF15 receptor agonist. It was designed to reduce appetite and promote weight loss by activating the GFRAL–RET receptor complex in the hindbrain, which mediates the anorexigenic effects of GDF15. In preclinical studies, MBL949 led to significant reductions in food intake and body weight across multiple animal species, with improvements in metabolic endpoints primarily due to fat loss. In clinical trials involving overweight or obese adults (with or without type 2 diabetes), subcutaneous administration of MBL949 every two weeks for up to 14 weeks resulted in minimal weight loss compared to placebo and did not demonstrate sufficient efficacy for further development. The most common adverse events were gastrointestinal-related, but overall safety was acceptable[4][5][2].
