MBP346 is a novel small molecule pyrazole pyrimidine derivative currently in preclinical development for the treatment of head and neck squamous cell carcinoma (HNSCC). It functions by inducing the production of reactive oxygen species (ROS), which leads to a reduction in mitochondrial membrane potential and triggers mitochondrial-mediated apoptosis. Additionally, MBP346 inhibits cell proliferation by inducing S-phase cell cycle arrest. This arrest is associated with the downregulation of key cell cycle regulators, including Cyclin D1, Cyclin A2, and CDK2, and the upregulation of the cyclin-dependent kinase inhibitor P21. In vitro studies have shown that MBP346 significantly inhibits the growth of HNSCC cell lines like Cal33 and Scc15 while exhibiting lower toxicity toward normal oral keratinocytes.
