MBRC-101 is a novel antibody-drug conjugate (ADC) that targets EphA5, a transmembrane tyrosine kinase receptor highly expressed in various malignant solid tumors including non-small cell lung cancer (NSCLC), triple negative breast cancer (TNBC), colorectal, gastric, pancreatic, and hepatocellular cancers. The ADC consists of an anti-EphA5 monoclonal antibody conjugated to the cytotoxic payload monomethyl auristatin E (MMAE). Upon binding to EphA5 on cancer cells, MBRC-101 is internalized and releases MMAE intracellularly to disrupt microtubule function leading to cancer cell death. This targeted approach aims to provide effective treatment with potentially reduced side effects compared to conventional chemotherapy. Developed by MBrace Therapeutics, MBRC-101 is currently in clinical development for advanced refractory solid tumors.
