MBS301 is a glyco-engineered, fully afucosylated bispecific monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2). It is constructed by combining two half antibodies derived from trastuzumab and pertuzumab, enabling simultaneous binding to two distinct epitopes on HER2. This biparatopic design enhances antitumor activity through improved antibody-dependent cell cytotoxicity (ADCC) and T lymphocyte stimulation. The afucosylation increases its affinity for FcγRIIIa receptors on immune effector cells, further boosting ADCC against HER2-positive cancer cells. Developed by Beijing Mabworks Biotech, MBS301 is being investigated primarily for the treatment of HER2-positive solid tumors that are resistant to trastuzumab[1][3][4][5].
