Overview

MBT-0312 (also known as EndoTAG-2) is an investigational cationic liposomal formulation of the cytostatic drug docetaxel, developed by Medigene (originally Munich Biotech). The therapy utilizes the proprietary EndoTAG platform, which consists of positively charged lipid complexes that encapsulate docetaxel. These cationic liposomes are designed to selectively target and bind to the negatively charged activated endothelial cells of newly forming tumor blood vessels (neoangiogenesis). By delivering docetaxel directly to the tumor vasculature, MBT-0312 acts as a vascular disrupting agent to destroy the tumor's blood supply, effectively "starving out" the tumor. This mechanism is intended to complement traditional anti-angiogenic strategies. Development of MBT-0312 was primarily focused on solid tumors but was discontinued as Medigene shifted its strategic focus toward T-cell receptor-based immunotherapies.
