MBX-2044 is a small molecule drug developed as an oral insulin sensitizer for the treatment of type 2 diabetes. It acts as a modulator of peroxisome proliferator-activated receptor gamma (PPARγ), aiming to improve insulin sensitivity and lower blood glucose levels. Unlike traditional thiazolidinediones (TZDs), which are full PPARγ agonists and associated with side effects such as weight gain and edema, MBX-2044 was designed to offer similar therapeutic benefits with an improved safety profile. The drug reached Phase 2 clinical trials but development was discontinued before approval[1][3][5]. Originally developed by Metabolex (later CymaBay Therapeutics), it was also subject to licensing agreements involving Ortho-McNeil (a Johnson & Johnson company) and later involved Janssen Global Services[1][3][8].
