Overview

MBX 5765 is a preclinical-stage amycretin prodrug development candidate being developed by MBX Biosciences for the treatment of obesity. The drug is a single-construct peptide therapeutic that combines agonist activity at the glucagon-like peptide-1 (GLP-1) receptor, glucose-dependent insulinotropic polypeptide (GIP) receptor, and glucagon (GCG) receptor, as well as dual amylin and calcitonin receptor agonist (DACRA) activity. This multi-targeted approach is enabled by the company's proprietary Precision Endocrine Peptide (PEP) platform, which allows for the creation of long-acting prodrugs. MBX 5765 is designed to offer superior weight loss efficacy and improved tolerability compared to existing incretin therapies, with a pharmacokinetic profile supporting once-monthly subcutaneous dosing. IND-enabling studies for the candidate are expected to begin in the second quarter of 2026.
