MC-100093 is a novel synthetic monocyclic beta-lactam (monobactam) derivative that acts as a potent enhancer of glutamate transporter 1 (GLT-1). Unlike traditional beta-lactam antibiotics such as ceftriaxone, MC-100093 is specifically designed to lack antimicrobial properties, thereby eliminating the risk of bacterial resistance during chronic administration. It possesses superior drug-like characteristics, including enhanced blood-brain barrier permeability and oral bioavailability. Preclinical research demonstrates that MC-100093 upregulates the expression and functional activity of GLT-1 and the cystine/glutamate antiporter (xCT) in mesocorticolimbic brain regions, which helps restore glutamate homeostasis disrupted by chronic substance exposure. The compound has shown efficacy in reducing the reinstatement of drug-seeking behaviors for cocaine, alcohol, nicotine, and opioids, and it also exhibits neuroprotective, cardioprotective, and hepatoprotective effects by mitigating neuroinflammation and oxidative stress.
