MC-230078 is a highly selective small molecule antagonist of the 5-hydroxytryptamine receptor type 7 (5-HT7). Developed by researchers at McMaster University, it is being investigated as a potential therapeutic for inflammatory bowel disease (IBD), including Crohn's disease and ulcerative colitis. Serotonin (5-HT) signaling is frequently altered in chronic gastrointestinal inflammatory disorders, and the 5-HT7 receptor subtype has been identified as a key mediator of mucosal inflammation. In preclinical murine models of colitis, including dextran sulfate sodium (DSS)-induced and T-cell transfer models, administration of MC-230078 significantly reduced intestinal inflammation, histopathological damage, and the production of proinflammatory cytokines compared to vehicle-treated controls.
