MC-30 is an investigational antibody-RNA conjugate (ARC) developed by miRecule for the treatment of head and neck squamous cell carcinoma (HNSCC). It consists of the anti-EGFR monoclonal antibody cetuximab conjugated to a chemically modified mimic of the tumor suppressor microRNA miR-30-5p. MC-30 utilizes a three-pronged mechanism of action: the cetuximab component binds to EGFR on the cell surface to inhibit downstream signaling and induce antibody-dependent cellular cytotoxicity (ADCC), while also facilitating receptor-mediated internalization of the conjugate. Once inside the cell, the miR-30-5p mimic payload is released to simultaneously downregulate multiple key oncogenic growth factor receptors, including EGFR, MET, and IGF-1R, thereby overcoming resistance mechanisms commonly associated with single-agent EGFR inhibition.
