MC-4 is a partially purified material derived from the plant *Artemisia annua* L. (sweet wormwood) that is being investigated for its potential as an anticancer therapy, specifically for advanced renal cell carcinoma (RCC). It functions as a novel inhibitor of the Akt and pyruvate kinase muscle isozyme M2 (PKM2) pathways. By enhancing the expression of the tumor suppressor phosphatase and tensin homolog (PTEN), MC-4 reduces downstream Akt/PKM2 signaling, which in turn decreases the expression of glucose transporter 1 (GLUT1) and disrupts cancer cell metabolism. Preclinical research indicates that MC-4 monotherapy induces autophagic cell death and tumor growth inhibition. Furthermore, when used in combination with the mTORC1 inhibitor everolimus, MC-4 demonstrates synergistic antitumor and anti-metastatic effects by overcoming compensatory Akt activation typically induced by mTOR inhibition.
