Overview

MC-45308 is a potent and selective small-molecule inhibitor of Myeloid Cell Leukemia-1 (MCL-1), a key anti-apoptotic member of the BCL-2 protein family. MCL-1 is frequently upregulated in various cancers, particularly hematological malignancies like multiple myeloma and acute myeloid leukemia, where it sequesters pro-apoptotic proteins to prevent programmed cell death. MC-45308 functions as a BH3 mimetic, binding to the hydrophobic groove of MCL-1 and displacing pro-apoptotic proteins such as BIM, thereby activating the intrinsic apoptotic pathway. Developed by Medi-Can Pharmaceuticals, MC-45308 is designed to address therapeutic resistance in cancers that depend on MCL-1 for survival, offering a targeted approach to induce tumor cell death while minimizing off-target effects on other BCL-2 family members.
