MC9999 is a second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target Programmed Death-Ligand 1 (PD-L1). Developed by researchers at the Mayo Clinic, this therapy utilizes a humanized anti-PD-L1 monoclonal antibody to engineer T cells that can recognize and eliminate PD-L1-expressing cells. Unlike many CAR T therapies that focus solely on tumor antigens, MC9999 is designed to target both the tumor cells themselves and the immunosuppressive cells within the tumor microenvironment (TME), such as M2 macrophages, microglia, and tumor-associated macrophages (TAMs). Preclinical studies have demonstrated its efficacy in models of breast cancer, non-small cell lung cancer, melanoma, and glioblastoma, including the ability to penetrate the blood-brain barrier in intracranial tumor models.
