Overview

mCALR DAC is a preclinical-stage mutated calreticulin (mCALR)-targeted degrader antibody conjugate (DAC) being developed by Prelude Therapeutics for the treatment of CALR-mutated myeloproliferative neoplasms (MPNs), including essential thrombocythemia (ET) and myelofibrosis (MF). The therapeutic approach utilizes a monoclonal antibody targeting mutated calreticulin, a neoantigen selectively presented on the cell surface of malignant myeloid cells, conjugated to a proprietary targeted protein degradation payload (such as a CDK9 degrader or SMARCA2/4 degrader). By delivering the degrader payload directly to mCALR-positive cells, the conjugate selectively eliminates malignant clones while sparing healthy hematopoietic cells, offering a potentially disease-modifying therapy with an improved therapeutic index.
