MCAM CAR NK cells are an investigational cell therapy consisting of natural killer (NK) cells engineered to express a chimeric antigen receptor (CAR) targeting the Melanoma Cell Adhesion Molecule (MCAM), also known as CD146. MCAM is a cell surface glycoprotein highly expressed in various pediatric solid tumors, including osteosarcoma (OSA), and is associated with tumor progression and therapy resistance in the tumor microenvironment. These CAR NK cells are typically produced by expanding peripheral blood mononuclear cells (PBMCs) and introducing the CAR construct via non-viral electroporation. Preclinical data indicates that MCAM CAR NK cells exhibit potent anti-tumor activity against OSA xenografts, which can be further augmented by co-administration with anti-GD2 antibodies like dinutuximab to trigger antibody-dependent cellular cytotoxicity (ADCC).
