Drug intelligence / Profile preview
MCARH125 + MCARH109 is an investigational combination regimen consisting of two autologous chimeric antigen receptor (CAR) T-cell therapies: MCARH125, which targets B-cell maturation antigen (BCMA), and MCARH109, which targets G-protein coupled receptor class C group 5 member D (GPRC5D). Developed by Memorial Sloan Kettering Cancer Center (MSK) in collaboration with Eureka Therapeutics and Juno Therapeutics (a subsidiary of Bristol Myers Squibb), this dual-targeting cell therapy is designed to treat relapsed or refractory multiple myeloma. By simultaneously targeting BCMA and GPRC5D, the combination aims to overcome tumor heterogeneity and prevent single-antigen escape, which are common mechanisms of resistance in myeloma. The therapies are manufactured from a single apheresis and administered sequentially following lymphodepleting conditioning chemotherapy.
