Overview

MCC-950 (also known as CP-456773 or CRID3) is a potent and highly selective small-molecule inhibitor of the NLRP3 (NOD-, LRR-, and pyrin domain-containing protein 3) inflammasome. It functions by directly binding to the NACHT domain of NLRP3, thereby inhibiting its ATPase activity and preventing the conformational change required for inflammasome assembly and activation. This inhibition blocks the subsequent activation of caspase-1 and the processing of pro-inflammatory cytokines, specifically IL-1β and IL-18, as well as the induction of pyroptotic cell death. Originally identified by Pfizer and later extensively characterized by researchers at the University of Queensland and Trinity College Dublin, MCC-950 has become a gold-standard pharmacological tool for studying NLRP3-mediated inflammation. While MCC-950 itself has primarily served as a research reagent due to its profile in early studies, its discovery led to the development of clinical-stage NLRP3 inhibitors by companies such as Inflazome, which was subsequently acquired by Roche.
