Overview

mcFOXM1 is a non-integrating minicircle DNA gene therapy designed to deliver the human FOXM1 (Forkhead Box M1) cDNA to the pulmonary endothelium. FOXM1 is a pro-proliferative forkhead transcription factor that is activated following injury to promote endothelial repair and is essential for embryogenesis. Unlike traditional plasmids, mcFOXM1 is constructed without prokaryotic backbones, which reduces the risk of inducing inflammation and enhances its clinical compatibility for potential FDA approval. In preclinical models of severe Bronchopulmonary Dysplasia (BPD), mcFOXM1 delivered via nanoparticles has demonstrated the ability to expand the pulmonary endothelium, increase Ccnd1 expression, attenuate alveolar damage, and restore endomucin expression, suggesting its potential as a therapeutic for neonatal lung disorders characterized by vascular and alveolar injury.
