MCL-1 CPAB is a preclinical-stage cell-penetrating Alphabody (CPAB) developed by Complix in collaboration with VIB. It is designed to target the intracellular anti-apoptotic protein MCL-1 (Induced myeloid leukemia cell differentiation protein Mcl-1), which is frequently upregulated in various cancers and contributes to tumor cell survival and resistance to chemotherapy. Unlike traditional monoclonal antibodies that are generally limited to extracellular targets, the CPAB platform utilizes a small, stable protein scaffold (Alphabody) engineered to cross the cell membrane and bind to its target within the cytoplasm. By binding to the BH3-binding groove of MCL-1, the drug inhibits its interaction with pro-apoptotic proteins, thereby triggering programmed cell death (apoptosis) in tumor cells. This molecule serves as a proof-of-concept for Complix's ability to address high-value intracellular protein-protein interaction targets.
