Overview

MCLA-145 is a bispecific monoclonal antibody developed using Merus' proprietary Biclonics technology. It targets both Programmed death-ligand 1 (PD-L1) and Tumor necrosis factor receptor superfamily member 9 (CD137, also known as 4-1BB), functioning as a conditional CD137 agonist and PD-L1 inhibitor. By binding to CD137, MCLA-145 provides T-cell co-stimulation, enhancing anti-tumor immune responses. Simultaneously, it blocks the interaction between PD-L1 and its receptor PD-1 on T cells, thereby preventing immune suppression within the tumor microenvironment. This dual mechanism is designed to intensify antigen-specific T cell responses against tumors while limiting systemic toxicity by confining activity to the tumor site where both targets are present. MCLA-145 is being developed primarily for advanced or metastatic solid tumors and B-cell lymphomas.
