mCM10-L is an mRNA-lipid nanoparticle (LNP) cancer vaccine designed to target the α3 domain of MHC class I polypeptide-related sequence A (MICA) and MHC class I polypeptide-related sequence B (MICB). Developed in collaboration with ProMab Bio, the vaccine utilizes an LNP delivery system (comprising SM-102, DSPC, cholesterol, and DMG-PEG2000) to deliver codon-optimized mRNA encoding MICA/B antigens. Its primary mechanism of action is to induce the production of specific antibodies that bind to the MICA/B α3 domain, thereby preventing the proteolytic shedding of these ligands from the tumor cell surface. By maintaining MICA/B expression on tumor cells and reducing levels of immunosuppressive soluble MICA/B (sMICA/B), the vaccine enhances the cytotoxic activity of NK cells and CD8+ T cells. Preclinical studies have demonstrated its efficacy in preventing metastasis and treating established metastatic disease in models of colorectal cancer, melanoma, and breast cancer.
