Overview

MCM998 is an autologous chimeric antigen receptor T-cell (CAR-T) therapy targeting B-cell maturation antigen (BCMA), developed by Novartis for the treatment of multiple myeloma. The therapy utilizes a second-generation CAR construct featuring an anti-BCMA single-chain variable fragment (scFv) fused to a 4-1BB (CD137) costimulatory domain and a CD3ζ signaling domain. MCM998 was investigated in early clinical trials, such as NCT03302403, where it demonstrated clinical activity in patients with relapsed or refractory multiple myeloma. In later development, Novartis has utilized MCM998 as a clinically validated benchmark construct to evaluate the performance of its next-generation CAR-T platform, T-Charge (e.g., PHE885), which aims to improve T-cell stemness and manufacturing efficiency.
