Overview

MCMV-TRP2 is a viral vector-based cancer vaccine consisting of a murine cytomegalovirus (MCMV) engineered to express the melanoma-associated antigen tyrosinase-related protein 2 (TRP2). Developed by researchers at Oregon Health and Science University, the vaccine is designed to induce anti-tumor immunity against melanoma. In preclinical mouse models (B16 melanoma), vaccination with MCMV-TRP2 demonstrated both prophylactic and therapeutic efficacy, significantly delaying tumor onset and prolonging survival. Unlike many other melanoma vaccines that rely on CD8+ T-cell responses, the mechanism of action for MCMV-TRP2 is primarily mediated by the induction of long-lasting TRP2-specific antibodies (humoral immunity). A spread-deficient version lacking the essential glycoprotein gL (ΔgL-MCMV-TRP2) has also been evaluated, showing similar anti-tumor effects while offering a potentially safer profile.
