The MCP-1-based GAGbody (also known as ATG01 or dnCCL2-HSA) is a recombinant fusion protein developed by Antagonis Biotherapeutics for the treatment of cancer metastasis. It consists of a dominant-negative mutant of monocyte chemoattractant protein-1 (MCP-1/CCL2) fused to human serum albumin (HSA) to extend its circulating half-life. The drug functions as a glycan antagonist by binding with high affinity to glycosaminoglycans (GAGs) on the vascular endothelium, thereby displacing native CCL2 and disrupting the chemokine gradients required for the recruitment of inflammatory monocytes and tumor cell extravasation. Additionally, the mutations in the CCL2 moiety prevent the activation of the CCR2 receptor, further inhibiting the pro-metastatic signaling axis. Preclinical studies have demonstrated its ability to reduce pulmonary metastatic burden in murine models.
