Overview

MCP-1 neutralizing antibody refers to a class of monoclonal antibodies designed to bind and inhibit Monocyte Chemoattractant Protein-1 (MCP-1), also known as CCL2. CCL2 is a key chemokine involved in the recruitment of monocytes, memory T cells, and dendritic cells to sites of inflammation and within the tumor microenvironment. In oncology, particularly in glioblastoma, CCL2 is secreted by tumor cells to recruit microglia and macrophages, which then promote tumor proliferation and invasion. By neutralizing CCL2, these antibodies aim to disrupt this recruitment and the subsequent pro-tumorigenic signaling. Clinical candidates in this class, such as carlumab (CNTO 888), have been investigated in various solid tumors and inflammatory conditions, though they have generally struggled to demonstrate significant clinical benefit as monotherapies, possibly due to compensatory mechanisms or rapid ligand turnover.
