MCS110 + PDR001 is a combination of two investigational monoclonal antibodies developed for the treatment of advanced solid tumors. Lacnotuzumab (MCS110) is a high-affinity, humanized monoclonal antibody targeting colony-stimulating factor 1 (CSF-1), which blocks the interaction of CSF-1 with its receptor on tumor-associated macrophages (TAMs), aiming to reduce TAM-mediated resistance to immunotherapy. Spartalizumab (PDR001) is a high-affinity, ligand-blocking, humanized anti–PD-1 immunoglobulin G4 antibody that inhibits the binding of PD-L1 and PD-L2 to PD-1 on T cells, thereby enhancing antitumor immune responses. The rationale for combining these agents is to overcome resistance mechanisms in tumors by depleting TAMs and simultaneously blocking immune checkpoints[1][2][3][5].
