The MCSP BiTE antibody is a preclinical bispecific T-cell engager (BiTE) originally developed by Micromet (now Amgen). It is designed to target melanoma-associated chondroitin sulfate proteoglycan (MCSP), also known as CSPG4, which is highly expressed on the surface of most human melanoma cells. The antibody construct simultaneously binds to MCSP on tumor cells and the CD3 subunit of the T-cell receptor complex on T cells. This dual binding facilitates the formation of a cytolytic synapse between the T cell and the tumor cell, leading to T-cell activation and the redirected lysis of the MCSP-expressing cancer cell. Preclinical data has demonstrated potent, dose-dependent anti-tumor activity against melanoma cell lines using both healthy donor and patient-derived T cells. The molecule was part of a next-generation BiTE platform designed to be cross-reactive with non-human primate species for improved safety assessment.
