Overview

MCU-i4 is a small molecule inhibitor of the mitochondrial calcium uniporter (MCU), a highly selective calcium channel located in the inner mitochondrial membrane. MCU facilitates the transport of calcium ions from the cytosol into the mitochondrial matrix, where moderate calcium levels stimulate ATP production and cell growth. In various cancers, including breast cancer, MCU is frequently overexpressed, leading to enhanced mitochondrial calcium uptake that supports rapid cancer cell proliferation and metabolic demands. By inhibiting MCU, MCU-i4 reduces mitochondrial calcium levels, thereby suppressing ATP production and inducing cell death. Research has specifically demonstrated its efficacy in BT474 breast cancer cell lines, highlighting its potential as a targeted antineoplastic agent.
