MD-164 is a small molecule inhibitor of the Arp2/3 complex, specifically targeting the ARPC2 (subunit 2) component. Developed by researchers at the Korea Research Institute of Bioscience and Biotechnology (KRIBB), it was identified through a screen of pharmaceuticals and preclinical drugs for inhibitors of tumor cell migration. By modulating the Arp2/3 complex, which is essential for actin polymerization and the formation of lamellipodia and invadopodia, MD-164 effectively reduces the migration and invasion of various cancer cell lines. In preclinical studies, it has demonstrated significant anti-metastatic activity, including a 70% reduction in lung metastases in a pancreatic cancer mouse model.
