Overview

MDC-250 is an allogeneic macrophage-drug conjugate (MDC) being developed by Cellis for the treatment of solid tumors, with a primary focus on pancreatic cancer. The MDC platform utilizes macrophages as active delivery vehicles for ferritin-encapsulated anti-cancer payloads. Macrophages internalize human heavy chain ferritin (HFt) via the macrophage scavenger receptor 1 (MSR1). Once at the tumor site, these drug-loaded macrophages transfer the ferritin-drug complexes directly to cancer cells through a novel mechanism termed TRAnsfer of Iron-binding proteiN (TRAIN). This process requires direct cell-to-cell contact and the formation of an immune synapse-like structure, enabling precise drug delivery. MDC-250 has demonstrated potent anti-tumor activity in preclinical orthotopic human pancreatic cancer models (BxPC3) and is designed as an off-the-shelf, cryopreserved product.
