MDC1 shRNA is a short hairpin RNA designed to silence the expression of the Mediator of DNA Damage Checkpoint 1 (MDC1) protein. MDC1 is a critical adaptor protein in the ataxia telangiectasia mutated (ATM) signaling pathway, responsible for the recruitment and retention of DNA repair factors at sites of double-strand breaks. In cervical cancer cells, high MDC1 expression is associated with resistance to chemotherapy and radiotherapy. By knocking down MDC1, this shRNA sensitizes cancer cells to genotoxic stress, leading to increased apoptosis and reduced DNA repair capacity. Research conducted at the Indian Institute of Technology-Gandhinagar has demonstrated that stable knockdown of MDC1 in cervical cancer cell lines (HeLa, SiHa, CasKi) significantly enhances the efficacy of DNA-damaging treatments.
