Overview

MDG1014 is a T-cell receptor (TCR) engineered T-cell therapy candidate targeting the cancer/testis antigen PRAME (Preferentially expressed Antigen in Melanoma). Developed by Medigene, the therapy utilizes a high-affinity, specific TCR that recognizes a PRAME-derived peptide presented by the HLA-A*02:01 molecule. MDG1014 is designed for the treatment of various solid tumors, including melanoma, where PRAME is highly expressed. To overcome the immunosuppressive tumor microenvironment, the TCR can be co-expressed with a PD1-41BB switch receptor, which converts inhibitory PD-1/PD-L1 signaling into stimulatory 4-1BB signaling, thereby enhancing T-cell fitness and anti-tumor activity.
