Overview

MDG10xx is a preclinical T cell receptor engineered T cell (TCR-T) therapy program from Medigene that comprises multiple candidate TCR-T therapies against several undisclosed solid-tumor targets and is designed to be combined with Medigene’s PD1-41BB switch receptor to enhance T cell activation and function in immunosuppressive tumor microenvironments.[1][3] In 2022, Medigene disclosed that BioNTech would acquire a next-generation preclinical TCR program using TCR-4 from Medigene’s MDG10xx program targeting PRAME together with the PD1-41BB switch receptor technology, while Medigene continues broader MDG10xx discovery against additional targets.[4][5][6] Mechanistically, the PD1-41BB switch receptor replaces the inhibitory PD-1 intracellular domain with an activating 4-1BB domain so that PD-L1 engagement delivers a costimulatory signal, supporting improved proliferation, repeated tumor cell killing, and metabolic fitness of TCR-T cells.[4]
