Overview

MDG2011 is a third-generation T cell receptor engineered T cell (TCR-T) therapy developed by Medigene. It specifically targets the KRAS G12V mutation presented on human leukocyte antigen HLA-A*11, a neoantigen commonly found in various solid tumors. The therapy incorporates Medigene’s proprietary PD1-41BB costimulatory switch protein (CSP) technology to enhance and armor the TCR-T cells, enabling them to overcome immunosuppressive signals within the tumor microenvironment. This approach aims to improve specificity, sensitivity, and safety of TCR-T therapies for difficult-to-treat solid tumors by combining optimal affinity mutant KRAS-specific TCRs with advanced costimulatory signaling[1][4][6]. Preclinical data have demonstrated high specificity and efficacy in targeting mKRAS G12V-HLA-A*11 positive tumor cells[2][7].
