MDG2021 is a T cell receptor engineered T cell (TCR-T) therapy developed by Medigene AG. It specifically targets the KRAS G12D mutation presented by human leukocyte antigen HLA-A*11, a common neoantigen in various solid tumors, including pancreatic adenocarcinoma. The therapy utilizes an optimal affinity 3S (sensitive, specific, and safe) TCR to recognize and eliminate tumor cells expressing this mutation while minimizing off-target effects on healthy tissues. MDG2021 is being developed both as a standalone TCR-T product and in combination with Medigene’s proprietary PD1-41BB costimulatory switch protein (CSP) technology to enhance anti-tumor efficacy by overcoming immunosuppressive mechanisms within the tumor microenvironment. As of June 2024, it is in preclinical development[2][3][5][6].
