Overview

MDH-1 inhibitor is a small molecule drug candidate being developed by the Global Alliance for TB Drug Development (TB Alliance) for the treatment of tuberculosis (TB). The drug targets Malate Dehydrogenase 1 (MDH-1), an essential enzyme in the citric acid cycle (TCA cycle) of *Mycobacterium tuberculosis*. MDH-1 catalyzes the reversible oxidation of malate to oxaloacetate, a critical step for bacterial energy metabolism and biosynthetic pathways. By inhibiting this enzyme, the drug aims to disrupt the metabolic viability of the pathogen, potentially providing a new therapeutic avenue against both drug-sensitive and drug-resistant TB strains. The program is currently in the lead identification and preclinical stage of development.
