Overview

MDI-0151 is a novel small molecule inhibitor of cathepsin C (also known as dipeptidyl peptidase I or DPP-1), developed initially by Modulus Discovery and further advanced by Alivexis. It acts as a non-covalent inhibitor targeting cathepsin C, an enzyme responsible for activating proteases involved in tissue destruction mediated by neutrophils. By inhibiting cathepsin C, MDI-0151 aims to reduce excessive neutrophil activation implicated in various inflammatory and immune-mediated diseases without significantly compromising overall immune function. The drug is being developed for refractory inflammatory conditions driven by increased neutrophil activity, such as ANCA-associated vasculitis and potentially other diseases including bronchiectasis, chronic obstructive pulmonary disease (COPD), hidradenitis suppurativa, palmoplantar pustulosis, inflammatory bowel disease (IBD), and systemic lupus erythematosus. Melodia Therapeutics has obtained exclusive global rights to develop and commercialize the compound from Alivexis[1][2][3][4][6].
