MDK-1654 is a branched synthetic peptide developed by Medikine, Inc., utilizing its proprietary PEPTIKINE technology platform. It functions as a dual-acting agonist that simultaneously activates the interleukin-7 (IL-7) receptor and the non-alpha (βγc) form of the interleukin-2/15 (IL-2/15) receptor. By mimicking the activity of these cytokines, MDK-1654 is designed to stimulate both the adaptive and innate immune systems, specifically promoting the survival, proliferation, and activation of T cells and natural killer (NK) cells. This dual mechanism aims to drive potent anti-tumor responses while potentially avoiding the regulatory T cell activation and toxicity associated with traditional IL-2 therapies that bind the alpha receptor subunit. Preclinical proof-of-concept data were presented at the Society for Immunotherapy of Cancer (SITC) 2022 meeting, supporting its development for oncology indications.
