Overview

MDK-271 is a preclinical **dual-function bispecific Fc fusion protein** being developed by Medikine for **cancer immunotherapy**. The molecule is a heterodimeric biologic built using knob-in-hole Fc engineering and contains one copy each of MDK1169, an **IL-2/15 receptor beta gamma c biased agonist**, and MDK1319, an **IL-7 receptor agonist**. By co-engaging cytokine receptor pathways involved in lymphocyte survival, expansion, and activation, MDK-271 drives STAT5 phosphorylation and has shown additive, complementary, and synergistic effects across multiple immune cell subsets, including CD4 positive T cells, CD8 positive T cells, regulatory T cells, memory T cells, and natural killer cells. The program is intended to enhance antitumor immunity through rational cytokine pathway co-stimulation rather than conventional monoclonal antibody blockade.
