Overview

MDK1472 is a synthetic 42-amino acid peptide that acts as a potent agonist of the interleukin-7 receptor (IL-7R). Developed by Medikine, Inc., it was discovered through phage display and is structurally unrelated to native IL-7, binding to the IL-7Rα (CD127) and common gamma chain (γc, CD132) subunits at sites distinct from the natural cytokine. MDK1472 functions as a full agonist, inducing STAT5 signaling and promoting the survival and expansion of naive and memory T-cells, with a particularly robust effect on stem-like memory T-cells (Tscm). Due to its novel sequence, it is designed to avoid the induction of neutralizing antibodies against endogenous IL-7, a limitation observed with some recombinant IL-7 therapies. MDK1472 serves as the pharmacologically active component of MDK-703, an IgG2-Fc fusion protein designed for extended circulating half-life, which is currently being evaluated in clinical trials for the treatment of advanced solid tumors and other conditions such as lymphopenia and infectious diseases.
