MDL-101 is a novel small molecule interleukin-6 (IL-6) inhibitor. It functions by suppressing the IL-6/signal transducer and activator of transcription 3 (STAT-3) signaling pathway. This inhibition leads to the suppression of autoreactive Th17 cell development and proliferation, while promoting the development of regulatory T cells (Treg). In in vitro studies, MDL-101 demonstrated superior potency in suppressing IL-17 production in myelin-specific CD4 T cells compared to related compounds MDL-5 and MDL-16. It also reduced the encephalitogenic potential of these cells and promoted human Treg development. MDL-101 is being investigated as a potential therapeutic strategy for multiple sclerosis (MS) by shifting the effector T cell (Teff) to regulatory T cell (Treg) balance.
