Overview

MDL-4101 is a potent, selective, and orally bioavailable small molecule inhibitor of bromodomain-containing protein 4 (BRD4), discovered and developed by Model Medicines using their proprietary AI-driven GALILEO platform. BRD4 is a member of the bromodomain and extra-terminal (BET) protein family and acts as a key regulator of oncogene expression, driving tumorigenesis in multiple solid and hematologic malignancies such as breast cancer, prostate cancer, leukemias, lymphomas, thyroid cancer, glioblastoma, prostate cancer, and testicular cancers. MDL-4101 was designed to overcome the limitations of previous BRD4 inhibitors by achieving high selectivity and improved tolerability. Preclinical studies have demonstrated robust anti-tumor activity in models of aggressive human thyroid cancer (CGTH-W-1), with evidence for efficacy in other cancers as well. As of June 2024, MDL-4101 has entered Phase 1 clinical trials for thyroid cancer[1][2][3][5].
