MDL-72145 is a small molecule research compound that acts as a potent, selective, and irreversible inhibitor of monoamine oxidase type B (MAO-B). Developed by Marion Merrell Dow, it is an enzyme-activated "suicide" inhibitor designed to treat Parkinson's disease by augmenting L-DOPA therapy. Unlike first-generation MAO inhibitors, MDL-72145 does not produce the "cheese effect" (hypertensive crisis from tyramine) and lacks the amphetamine-like sympathomimetic metabolites associated with selegiline. Preclinical studies demonstrated its ability to prevent MPTP-induced neurotoxicity in animal models. It also exhibits inhibitory activity against semicarbazide-sensitive amine oxidase (SSAO). While it reached early clinical evaluation in healthy volunteers, it did not progress to late-stage clinical trials or commercial approval.
