Overview

MDM2 siRNA refers to small interfering RNA molecules designed to silence the expression of the MDM2 (Mouse double minute 2 homolog) gene. MDM2 is an E3 ubiquitin ligase that acts as the primary negative regulator of the p53 tumor suppressor protein by targeting it for proteasomal degradation. By utilizing the RNA interference (RNAi) pathway to reduce MDM2 protein levels, these siRNAs stabilize p53, leading to the activation of p53-dependent pathways such as cell cycle arrest and apoptosis. Preclinical research, including studies in testicular carcinoma, Hodgkin lymphoma, and acute myeloid leukemia, has demonstrated that MDM2 siRNA can significantly sensitize cancer cells to chemotherapy (e.g., cisplatin) by promoting apoptosis, often through the induction of the Fas/FasL death receptor pathway.
