MDMG-935P is a small molecule salicylic acid derivative that functions as a dual inhibitor of lactate dehydrogenase A (LDHA) and glycolate oxidase (HAO1/GO). It was developed to address hepatic oxalate overproduction, a metabolic pathway implicated in the progression of metabolic dysfunction-associated steatohepatitis (MASH). By inhibiting LDHA and HAO1, MDMG-935P prevents the accumulation of oxalate from glyoxylate in hepatocytes. Preclinical studies in mouse models of MASH demonstrate that MDMG-935P treatment can reduce hepatic lipid accumulation, enhance fatty acid β-oxidation, and suppress proinflammatory responses, including monocyte infiltration, thereby ameliorating disease severity.
