MDNA109FEAA-Fc-MDNA132 is a preclinical bi-functional SuperKine ImmunoTherapy (BiSKIT) developed by Medicenna Therapeutics. It is a recombinant fusion protein designed to deliver a potent interleukin-2 (IL-2) agonist specifically to the tumor microenvironment (TME). The molecule consists of MDNA109FEAA, an engineered "beta-only" IL-2 super-agonist that selectively activates the IL-2 receptor beta/gamma (IL-2Rβγ) complex without binding to the high-affinity alpha subunit (CD25), thereby avoiding the stimulation of regulatory T cells and vascular leak syndrome. This IL-2 moiety is fused via an Fc scaffold to MDNA132, an engineered IL-13 superkine with high specificity for the IL-13 decoy receptor (IL13Rα2) and no binding to the functional IL13Rα1. Since IL13Rα2 is highly overexpressed in various cancers—including glioblastoma, pancreatic, and colorectal cancer—while having limited expression in normal tissues, MDNA109FEAA-Fc-MDNA132 enables targeted immune activation within the TME. Preclinical studies have demonstrated that the compound maintains selective receptor binding, stimulates pSTAT5 signaling with high potency, and selectively localizes to IL13Rα2-expressing tumors.
